Does deathtouch work through trample when assigning combat damage?

Short answer
Yes. A deathtouch trampler only needs to assign 1 lethal damage to each blocker, then excess damage can be assigned to the defending player.

When a creature with both deathtouch and trample attacks and is blocked, the attacking player assigns combat damage. Normally, trample requires you to assign lethal damage to each blocker before assigning any damage to the defending player (CR 702.19b). Deathtouch modifies what counts as 'lethal': any amount of damage dealt by a source with deathtouch is considered lethal damage (CR 702.2b, 702.19c).

Because of this interaction, an attacker with deathtouch and trample only needs to assign 1 damage to each blocking creature (since even 1 damage is lethal), and all remaining damage can be assigned as trample damage to the defending player or planeswalker.

This is explicitly addressed in CR 702.19c: 'when a creature with trample also has deathtouch, the controller of that creature assigns combat damage by assigning 1 damage to each creature blocking it and the rest to the defending player or planeswalker.'

Example: A 6/6 creature with trample and deathtouch is blocked by two 5/5 creatures. Normally trample would require 10 damage assigned to blockers, leaving nothing for the player. With deathtouch, the attacker assigns 1 damage to each 5/5 (both are killed since any damage is lethal) and assigns the remaining 4 damage directly to the defending player.

This is a very powerful combination because large toughness blockers can no longer 'absorb' the entire attack — the deathtouch trampler efficiently kills blockers and still pushes damage through.
